Researchers uncover the molecular mechanisms leading to basal cell carcinoma initiation

One of the most outstanding and unresolved questions in cancer biology is the identification of cells at the origin of cancer and the understanding of the molecular changes that occur in tumor initiating cells from the first oncogenic mutation to the development of invasive cancer. Basal cell carcinoma (BCC) is the most frequent cancer in human with more than million of new patients diagnosed each year with BCCs. Recently, the group of Pr. Cédric Blanpain uncovered the cells at the origin of BCC (Youssef et al, Nature Cell Biology, 2010).

Tension on gut muscles induces cell invasion in zebrafish intestine, mimicking cancer metastasis

The stiffness of breast tissue is increasingly recognized as an important factor explaining the onset of breast cancer. Stiffening induces molecular changes that promote cancerous behavior in cells. Bioengineering studies have found that breast cancer cells grown on a 3-D gel have enhanced cell replication and decreased organization as rigidity increases. These signals are probably coordinated by surface proteins that communicate with connective tissue, to regulate cell replication, death, and movement. However, very little is known about how stiffness or other physical characteristics of tissues contributes to cancer behavior in living animals.

One in five women with breast cancer has a reoperation after breast conserving surgery

One in five women with breast cancer who opt for breast conserving surgery rather than a mastectomy have a reoperation, according to a national study published in the British Medical Journal today. This information on the risk of reoperation should help women in making the decision about whether to undergo breast conserving surgery or mastectomy.

Radiotherapy for DCIS still protects against recurrence after 15 years

Radiotherapy treatment (RT) after surgery for ductal carcinoma in situ (DCIS) still has a major protective effect against recurrence more than 15 years later, according to the results of an international trial. Researchers found that the use of RT in addition to surgery could reduce the chances of a local recurrence (the cancer coming back in the same breast) by 50%. Results from the trial, which has one of the longest follow-ups of a large group of patients in the world to date, will be reported today to the 8th European Breast Cancer Conference (EBCC-8).

Protein that functions in normal breast may also contribute to breast cancer metastasis

The trefoil factor 3 (TFF3) protein protects and maintains the integrity of the epithelial surface in the normal breast. New research has found that while TFF3 protein expression is higher in well-differentiated low grade tumors and therefore associated with features of a good prognosis, it has a more sinister role in breast cancer invasion and metastasis. The report is published in the March issue of the American Journal of Pathology.

How early breast tumors become deadly: A small group of molecules might hold the answer

Researchers have discovered a restricted pattern of molecules that differentiate early-stage breast tumors from invasive, life-threatening cancer. They also found a similar molecular signature that correlated with the aggressiveness of invasive tumors, and with the time to metastasis and overall survival.

A mutant protein found in nearly all pancreatic cancers plays a role not only in the cancer's development but in its continued growth, according to a new study from University of Michigan Comprehensive Cancer Center researchers. The finding suggests a possible target for developing new ways to treat this deadly disease.

Four-week vaccination regimen knocks out early breast cancer tumors, researchers find

Researchers at the Perelman School of Medicine at the University of Pennsylvania report that a short course of vaccination with an anti-HER2 dendritic cell vaccine made partly from the patient's own cells triggers a complete tumor eradication in nearly 20 percent of women with ductal carcinoma in situ (DCIS), an early breast cancer. More than 85 percent of patients appear to have a sustained immune response after vaccination, which may reduce their risk of developing a more invasive cancer in the future. The results of the study were published online this month of Cancer and in the January issue of the Journal of Immunotherapy.

Кesearchers examine 21-year series of nipple sparing mastectomy cases and find no cancers

A new study suggests some women needing a lumpectomy or mastectomy to treat their breast cancer have another potential option that is safe and effective, say researchers at Georgetown. They say the procedure known as a nipple sparing mastectomy is also a viable surgical option for women who choose to have their breasts removed because of their increased risk of developing the disease. For both groups of women, the surgery offers a chance for a more natural looking and normal feeling reconstructed breast as compared to other forms of mastectomy.

Promising new target for stifling the growth and spread of cancer

Cancer and chronic inflammation are partners in peril, with the latter increasing the likelihood that malignant tumors will develop, grow and spread. Researchers at the University of California, San Diego School of Medicine say they've identified a tumor inflammation trigger that is common to most, if not all, cancers. And using existing inhibitory drugs, the scientists were able to dramatically decrease primary tumor growth in animal studies and, more importantly, halt tumor progression and metastasis.
